
Popular GLP-1 class diabetes drugs may reduce the risk of serious bone fractures in people with type 2 diabetes, researchers reported in JAMA Network Open.
Over a three-year period, patients taking GLP-1 receptor agonists had a 21 percent lower risk of sustaining fractures caused by minor injuries compared to those taking DPP-4 inhibitors, another widely used class of diabetes drugs. The largest reduction in fracture risk was observed in the hip, femur, and spine.
Researchers analyzed medical records from nearly 134,000 Americans aged 50 to 90. The study evaluated outcomes for patients initiating GLP-1 receptor agonist therapy against a control group receiving DPP-4 inhibitors over the three-year tracking period.
Bone health concerns and study limitations
The findings ease prior concerns among medical experts that the rapid weight loss typically associated with GLP-1 therapy could weaken bone density and increase fracture risks. Bone density naturally declines with age, leaving older adults particularly susceptible to severe injury from minor falls. However, the study authors emphasized that their investigation was observational, meaning it does not prove a direct protective mechanism of GLP-1 drugs on bone tissue.
GLP-1 receptor agonists are a class of medications designed to mimic the glucagon-like peptide-1 hormone, which stimulates insulin release, lowers blood sugar levels, and promotes a feeling of fullness. DPP-4 inhibitors work by blocking the dipeptidyl peptidase-4 enzyme to prevent the degradation of natural GLP-1 hormones. Type 2 diabetes is a chronic metabolic condition where the body becomes resistant to insulin or fails to produce enough of it.
Broader research on GLP-1 treatments
The study adds to an expanding body of research surrounding the secondary health effects of GLP-1 medications. Scientists previously discovered that weight loss drugs are also associated with a reduced risk of death and cancer recurrence in patients diagnosed with breast cancer.
